Since its original evaluation by JECFA, titanium dioxide has become a public point of contention, with its ban being introduced (and then subsequently withdrawn) in California legislation in 2023, a legal battle playing out in the EU over the additive’s ban and classification as a carcinogen in 2022, and the European Food Safety Authority (EFSA) calling titanium dioxide unsafe. However, supporters of titanium dioxide say that claims about its dangers are founded in unreliable studies, and some recent research has supported its safety as a food additive.

On the other hand, Westerhoff said, there are hundreds of studies showing no adverse effects from the substance.

Assessment of skin penetration and biohazard in vivo

Because of health risks, France banned titanium dioxide as a food additive in 2020. Two years later the European Union also banned titanium dioxide as a food additive. 

Titanium dioxide has similar uses in non-food products. It is used in sunscreen as effective protection against UVA/UVB rays from the sun, which creates a physical barrier between the sun’s rays and the skin. It’s also used to whiten paint, paper, plastic, ink, rubber, and cosmetics.

Titanium dioxide exists primarily in three crystalline forms rutile, anatase, and brookite. Among these, anatase is characterized by its unique electronic properties and higher photocatalytic activity, making it suitable for applications requiring enhanced light absorption and reactivity. Its ability to act as a photocatalyst enables the breakdown of organic pollutants, making it a valuable component in environmental remediation strategies.

For a review published in 2023 in the journal Environmental Pollution, researchers examined E171 as a possible factor promoting obesity-related metabolic disorders. Because gut microbiota play an important role in immune function maintenance and development, and because titanium dioxide as a food additive has been shown to alter gut microbiota, researchers wanted to review “the dysregulations along the gut microbiota-immune system axis after oral TiO2 exposure compared to those reported in obese or diabetic patients, and to highlight potential mechanisms by which foodborne TiO2 nanoparticles may increase the susceptibility to develop obesity-related metabolic disorders.” The study authors discovered recurrent changes in the gut microbiota composition when exposed to titanium dioxide nanoparticles, with an imbalance of intestinal symbiotic microbiota. These changes and imbalances were also reported and played a role in the development of obesity, the authors wrote. This highlights “foodborne TiO2 nanoparticles as an endocrine disruptor-like chemical promoting obesity-related disorders,” the authors concluded.

Nanotoxicology “focuses on determining the adverse effects of nanomaterials on human health and the environment.”

For research published in Archives of Toxicology in 2020, scientists fed one group of mice a solution containing titanium dioxide for one month, and compared it to those that did not receive the additive. They found “the richness and evenness of gut microbiota were remarkably decreased and the gut microbial community compositions were significantly changed” in the titanium dioxide group when compared with the control group. The tests also revealed that the titanium dioxide exposure could cause locomotor dysfunction, or mobility issues “by elevating the excitement of enteric neurons, which might spread to the brain via gut-brain communication by vagal pathway.” The researchers concluded: “These findings provide valuable insights into the novel mechanism of TiO2NP-induced neurotoxicity. Understanding the microbiota-gut-brain axis will provide the foundation for potential therapeutic or prevention approaches against TiO2NP-induced gut and brain-related disorders.”

Freshwater algae show low-to-moderate susceptibility to TiO₂ exposure, with more pronounced toxic effects in the presence of UV irradiation. It has also been shown that nano-sized TiO₂ is significantly more toxic to algae Pseudokirchneriella sub-capitata than submicron-sized TiO₂. Hund-Rinke and Simon  reported that UV irradiated 25 nm TiO₂ NPs are more toxic to green freshwater algae Desmodesmus subspicatus than UV irradiated 50 nm particles, which is in agreement with Hartmann et al. UV irradiated TiO₂ NPs also inactivated other algae species such as Anabaena, Microcystis, Melsoira and Chroococcus. It was demonstrated that smaller particles have a greater potential to penetrate the cell interior than submicron-sized particles and larger aggregates. Studies have shown that the amount of TiO₂ adsorbed on algal cells can be up to 2.3 times their own weight.
